| Institution/Organization: Washington University Medical School in St. Louis Department Name: Internal Medicine/Renal Division Job Title:Postdoctoral Research Associate Field of Study:Renal Medicine Job Location:St. Louis Job Description:Two NIH-grant funded postdoctoral positions are available to investigate inherited renal diseases and the underlying molecular mechanisms. Our laboratory is interested in understanding the physiology of paracellular permeation of ions and solutes in the kidney. To this end, we have studied the genes mutated in human hypomagnesemia syndrome (FHHNC- Familial Hypomagnesemia with Hypercalciuria and Nephrocalcinosis), claudin-16 and claudin-19. Using lentiviral transgensis, we have generated transgenic mouse lines expressing siRNAs against claudin-16/-19 (Hou et al., J Biol Chem 2007, 282, 17114-17122). Using a number of genetic and biochemical analyses, we have revealed a selective protein interaction of claudin-16 with claudin-19 and that this claudin interaction underpins the pathogenesis of FHHNC (Hou et al., J Clin Invest. 2008, 118:619-28). Ongoing projects are focused on (1) defining the heteromeric channel properties of claudin-16/-19 complex; (2) studying the renal handling of electrolytes in claudin siRNA mice; (3) identifying novel claudins genetically linked to human salt-sensitive hypertension and hypochloremia. Our lab has established a number of cutting-edge techniques, including germ-line lentiviral transgenesis, virus mediated gene transfer; and sophisticated surgical methods for analyzing renal function. The renal division provides a highly interactive multidisciplinary research environment focused on translating basic science discoveries to the clinical therapies. Qualifications:MD or PhD with strong backgrounds in (1) biochemistry and protein interactions or (2) electrophysiology and animal physiology. No citizenship and permanent residency requirement.
